Data on the break-down of ATP during contraction of intact, rested muscle are contradictory, since they are limited by methodological difficulties in the study of shifts in the ADP-ATP system. The results of the work of Chans et al. are examined by differential spectrophotometry, using a two-beam spectrometer to study intact, unfrozen muscle. A description of the apparatus and method is given. In the intact muscle with intact respiratory enzymes capable of oxidation and reduction, ADP appears during contraction (in concentration of less than .001 uM per gram of muscle). The role of ADP as a regulator of intracellular metabolism is studied. Bibliography: 50 references.
